Orforglipron is a non-peptide, small-molecule agonist of the glucagon-like peptide-1 receptor (GLP-1R). Unlike peptide-based GLP-1 agonists, it is orally bioavailable without injection. It activates GLP-1R-mediated intracellular cAMP signalling to stimulate glucose-dependent insulin secretion, suppress glucagon, slow gastric emptying, and reduce appetite via hypothalamic pathways. Research applications include type 2 diabetes modelling, metabolic dysfunction studies, and oral GLP-1R pharmacology.
